As of 24 Mar 2016, COOL now accepts sequences which do not end with stop codons. However, if you exclude the stop codon, it will likewise be absent in the optimized results, which may cause translation runoff.
Protein Sample Protein Data: Human Insulin
Note: A valid protein sequence should start with Methionine (M), and consist of unambiguous amino acids (R, H, K, D, E, S, T, N, Q, C, G, P, A, V, I, L, M, F, Y, W). There should be a termination signal (*) at the end, but this is optional, and if a termination signal is excluded, it will likewise be absent in the optimized results.
 
DNA/RNA Sample DNA Data: Human Beta Actin
Note: A valid nucleotide sequence should start with Methionine (ATG/AUG), and must consist of unambiguous bases (A, C, G, T, U), and the number of bases should be divisible by 3. There should be a stop codon (e.g. TAA for Standard Code) at the end, but this is optional, and if an end stop codon is excluded, it will likewise be absent in the optimized results.
